Elisa evaluation of the levels of antibodies against Infectious Bronchitis Virus in laying hens using egg yolk as substrate Rev. Bras. Ciênc. Avic.
Rauber,RH; Flôres,ML; Pereira,CE; Grigulo,M.
The present work was carried out to compare Infectious Bronchitis Virus (IBV) antibody titers in serum and egg yolk samples from laying hens. Sixty paired blood and egg samples were collected from laying hens of two farms. Serum samples were frozen, while egg yolk samples were diluted (1:500) before freezing. Serum and yolk samples were tested for the presence of IBV antibodies by indirect ELISA (commercial kit) and titers were compared by a correlation test (alpha=0.05). There was a high correlation (r=0.62) between the two kinds of samples, which means that titers of IBV antibodies in the egg yolk and in serum samples are quite the same. Considering that blood collection causes deep stress that leads to economic losses, and since eggs are collected daily...

Experimental infection with Brazilian Newcastle disease virus strain in pigeons and chickens BJM
Carrasco,Adriano de Oliveira Torres; Seki,Meire Christina; Benevenute,Jyan Lucas; Ikeda,Priscila; Pinto,Aramis Augusto.
Abstract This study was designed with the goal of adding as much information as possible about the role of pigeons (Columba livia) and chickens (Gallus gallus) in Newcastle disease virus epidemiology. These species were submitted to direct experimental infection with Newcastle disease virus to evaluate interspecies transmission and virus-host relationships. The results obtained in four experimental models were analyzed by hemagglutination inhibition and reverse transcriptase polymerase chain reaction for detection of virus shedding. These techniques revealed that both avian species, when previously immunized with a low pathogenic Newcastle disease virus strain (LaSota), developed high antibody titers that significantly reduced virus shedding after...

Expression of Grapevine leafroll-associated virus 3 coat protein gene in Escherichia coli and production of polyclonal antibodies Trop. Plant Pathol.
Fajardo,Thor V.M.; Barros,Danielle R.; Nickel,Osmar; Kuhn,Gilmar B.; Zerbini,F. Murilo.
Grapevine leafroll-associated virus 3 (GLRaV-3), the main viral species of the grapevine leafroll complex, causes yield and quality reduction in grapes (Vitis spp.). The coat protein gene was RT-PCR-amplified from total RNA extracted from infected grapevine leaves and the amplified fragment was cloned and completely sequenced. The fragment was subsequently subcloned into the pRSET-C expression vector. The recombinant plasmid was used to transform Escherichia coli BL21:DE3 and express the capsid protein. The coat protein, fused to a 6 His-tag, was purified by affinity chromatography using an Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE and Western blot. The in vitro-expressed protein was quantified and used for rabbit...

First molecular and serological detection of Epizootic Hemorrhagic Disease virus in white tailed deer ( Odocoileus virginianus ) from Tamaulipas, Mexico Arq. Bras. Med. Vet. Zootec.
Merino,J.O.; Cruz,NI. De la; Galvan,G.; León,A.P. De; Burnes,J..
ABSTRACT Epizootic hemorrhagic disease viruses (EHDV) are dsRNA arboviruses transmitted by biting midges of the genus Culicoides that cause disease in domestic and wild ruminants. Epizootic hemorrhagic disease (EHD) is considered the most important infectious disease of white tailed deer (WTD) in North America, some studies in Northeast Mexico reported EHDV-seropositive WTD and EHDV-infected Culicoides vectors. The increasing population of WTD that share habitat with livestock in Northeast México highlights the importance of EHD for the livestock industry in the transboundary region with the U.S. One hundred and twenty two samples from WTD in Tamaulipas state, Mexico were tested by ELISA and RT-PCR for EHDV antibodies and nucleic acid, respectively. Twelve...

First survey for detecting the presence of human herpesvirus 8 infection (HHV-8) in Maputo, Mozambique BJID
Caterino-de-Araujo,Adele; Manuel,Rolanda Carmen Rafael; Bianco,Rosana Del; Santos-Fortuna,Elizabeth; Magri,Mariana Cavalheiro; Silva,Joyce Matie Kinoshita; Bastos,Rui.
Human herpesvirus 8 (HHV-8), the etiological agent of Kaposi's sarcoma (KS), is endemic in parts of the sub-Saharan, and KS has increased concomitantly with the HIV/AIDS epidemic. In Mozambique (MZ), no data concerning HHV-8 infection was available, thus the main of this work was to investigate, for the first time, the presence of HHV-8 infection in Maputo, MZ. Latent and lytic HHV-8-specific antibodies were assessed in blood samples from 189 individuals from the Central Hospital of Maputo, MZ, using "in-house" immunofluorescence assays conducted in São Paulo, Brazil. The results obtained were analyzed according to socio-demographic and clinical variables using the Chi-square test and logistic regression. An HHV-8 seropositivity of 1.8% and 9.7% was...

Genital schistosomiasis mansoni concomitant to genital tumor in areas of low endemicity: challenging diagnosis BJID
Cavalcanti,Marta G; Gonçalves,Margareth MI; Barreto,Magali M; Silva,Aline Helen da; Madi,Kalil; Peralta,José Mauro; Igreja,Ricardo P.
Genital infection by Schistosoma mansoni is usually misdiagnosed in individuals who reside in, or travel to endemic areas. We describe two cases of genital tumor associated with S. mansoni infection manifested by methrorragy. Surgical specimens revealed leiomyomas in both cases associated with S. mansoni. In one of them, granulomas were found in the ovary and in the other they were found in the uterine tube. Although none presented intestinal/hepatic disease, fecal egg excretion was detected in one. Both had elevated pretreatment antibody reactivity to S. mansoni antigen, but follow-up showed different outcomes. Schistosomiasis should be considered as a diagnosis in individuals with methrorragy residing in or having traveled to endemic areas. Since...

HUMORAL RESPONSE TO DIFFERENT VACCINATION SCHEMES AGAINST CLASSICAL SWINE FEVER (CSF) SUCCESSIVELY APPLIED DURING AN OUTBREAK OF THE DISEASE Rev Salud Anim.
Percedo,María Irian; Alfonso,P; Frías,María Teresa; Díaz de Arce,Heydi; Barrera,Maritza; Fonseca,O; Castell,Sara.
Vaccination against classical swine fever (CSF) is an essential measure for its control in endemic areas, and the Chinese Strain has been used with success in already disease free countries. Nevertheless the complexity of the factors interacting on the epidemiology and control of CSF, when the expected success is not reached with the vaccination of premises, doubts always fall on the vaccine used and the vaccination scheme applied In consecuences, many changes in vaccination scheme by the breeders are frecuently observed whitout positive influence in CSF control. To clarify the CSF recurrence in a herd vaccinated by a different scheme as the one recommended in the national control program, serological samples were taken to different animal groups in three...

Immunogenicity in sheep of Uruguayan commercial vaccines against bovine alphaherpesvirus 1, 5 and bovine pestiviruses Ciência Rural
Merchioratto,Ingryd; Aurélio,Alana de Almeida; Villela,Janice Machado; Stone,Nicole Vieira; Roman,Isac Junior; Traesel,Carolina Kist; Brum,Mário Celso Sperotto.
ABSTRACT: The serological responses induced by four commercial inactivated Uruguayan vaccines against bovine alphaherpesviruses (BoHV)-1 and -5 and bovine pestiviruses (BVDV-1, BVDV-2, and HoBiPeV) were evaluated in sheep. Thirty-seven sheep were immunized twice (day 0 and 25) and their serum samples were tested at different intervals (days 0, 25, 40, 60, and 90) post-vaccination (PV). Among the four vaccines tested, only one (G4) could induce the production of moderate neutralizing antibody titers against BoHV-1 and -5 and BVDV-1 and -2. The G3 vaccine showed a neutralizing serological response against the bovine alphaherpesviruses only. The G1 and G2 vaccines produced extremely low levels of antibodies in a few vaccinated animals only (geometric mean...

Improvement of an in-house ELISA for bovine paratuberculosis serology in Brazil BJM
Marassi,Carla Dray; Fonseca,Leila de Souza; Ristow,Paula; Ferreira,Rachel; Lilenbaum,Walter; Oelemann,Walter M.R..
Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Moreover, bacterial growth is slow and test results are available only after three to five months of incubation Furthermore, shedding of bacilli at levels detectable by fecal culture is irregular and does not occur during the early stages of infection, which compromises the sensitivity of this methodology. The most common immunological tests to identify Map infection are complement fixation test (CFT), agar gel immunodiffusion (AGID) and enzyme-linked immunoassay (ELISA). In Brazil,...

Scanavini Neto,H; Ito,NMK; Miyaji,CI; Lima,E de A; Okabayashi,S; Corrêa,ARA; Eleutério,GC; Zuanaze,MA.
IBDV Gm 11 (Simbios eleven-molecular group) has been detected since 1997 in many farms of commercial broilers and layers causing high mortality (2 to 15%) and severe macro and microscopic damage in cloacal bursae, spleen, thymus, kidney and liver. Five serial passages of 2050/97-Gm 11 IBDV sample by CAM route in SPF chicken's embryonated eggs did not elicit increased embryo mortality. High mortality (100%) of 21 day-old SPF leghorn chickens and severe bursal and splenic lesions were seen from 24 up to 48 hours after eye-drop inoculation of 2050/97 strain (50 mL of 10-2 dilution of 10% bursae homogenate). Mortality was not detected when vaccinated SPF and broiler chickens were inoculated. One dead bird was found among ten challenged unvaccinated broilers....

Latent human herpesvirus - 8 (HHV-8) infection in female commercial sex workers from Imbituba, Santa Catarina, Brazil BJID
Caterino-de-Araujo,Adele; Santos-Fortuna,Elizabeth; Magri,Mariana Cavalheiro; Schuelter-Trevisol,Fabiana; Silva,Marcos Vinicius.
Human herpesvirus 8 (HHV-8) infection was identified in 6 out of 90 (6.7%) female commercial sex workers from Imbituba, Santa Catarina, and was associated to age. Frequencies of 5.6% of anti-latent and 3.3% of anti-lytic antibodies were detected. Considering non-endemic areas from Brazil, the anti-latent antibodies frequency seems elevated and requires further investigation on referent female population.

Le concept de biosécurité en pisciculture marine : prévention de l'encéphalite virale chez l'ombrine subtropicale (Sciaenops ocellatus) en Martinique ArchiMer
Adee, Fabrice.
In 1985, red drum (Sciaenops ocellatus) was introduced in Martinique in arder to replace sea bass fish farming. Nodavirus antibodies were detected in the broodstock at the IFREMER station in Robert. Nodavirus is responsible for viral nervous necrosis, a disease that is spreading quickly in worlwide marine aquaculture. A biosecurity process was initiated at the Robert hatchery to select seronegative breeders and produce nodavirus free larvae. This process, based on a serological follow-up of fish antibodies by ELISA, allowed a reduction of the number of seropositive fish. A complementary serological study revealed a low incidence of nodavirus contamination in nodavirus free larvae placed in sea cages on different production sites

Mycoplasma synoviae infection on Newcastle disease vaccination of chickens BJM
Silva,Rita de Cássia Figueira; Nascimento,Elmiro Rosendo do; Pereira,Virgínia Léo de Almeida; Barreto,Maria Lúcia; Nascimento,Maria da Graça Fichel do.
Newcastle disease is characterized by respiratory manifestations in association with nervous and/or digestive symptoms. Its prevention is done by vaccination with live attenuated (lentogenic strains) and/or killed vaccines. The lentogenic strains can lead to strong post-vaccination reaction, principally due to the presence of other pathogenic agents. Among them, Mycoplasma synoviae is worldwide important, mainly in Brazil. The dissemination of this agent in poultry flocks has been achieved due to difficulties in diagnosis and disease reproduction, virulence variations among different M.synoviae strains, and attribution of typical M.synoviae disease manifestation to other disease agents. This experimental study in SPF chicks (Gallus gallus), previously...

Natural infection of Viola cornuta (Violaceae) with Cucumber mosaic virus, subgroup I Biocell
Arneodo,Joel; de Breuil,Soledad; Lenardon,Sergio; Conci,Luis.
Plants of Viola cornuta displaying typical virus symptoms were observed during spring 2003 in a plant nursery in Córdoba, central Argentina. Electron microscopic examinations of symptomatic leaf samples revealed the presence of isometric virus-like particles about 30 nm in diameter. Subsequent serological analysis allowed the identification of the pathogen as a subgroup I strain of Cucumber mosaic virus (CMV). These results were confirmed by antigen capture - reverse transcription - polymerase chain reaction with specific CMV primers, and digestion with a restriction enzyme. This is the first report of CMV infecting V. cornuta in Argentina.
